The Enterprise can go toe-to-toe with Klingon and Romulan battleships. It is intended to be an explorer in deep space that can fight off literally anything the unfriendly universe can throw at it, while still getting clear, usable scientific data. Which means tons of both defensive and offensive capabilities. It’s pretty much a Starfleet flagship, so anyone whose gunning for Starfleet will be gunning for the Enterprise.
The Millenium Falcon’s main attribute is that it can do the the Kessel Run in as short a time and using as many shortcuts as physically possible and still get its cargo safely delivered. The Falcon is fast, and anything that might slow it down (for example, extraneous weaponry or defensive capabilities) has probably been removed by Han with extreme prejudice.
